Fowley's acting career debuted in 1933 with "The Mad Game," alongside Spencer Tracy and Claire Trevor. He quickly made a name for himself, skillfully navigating through diverse roles. His most celebrated performance came in the 1952 musical Singin' in the Rain, where he played a film director. This role is emblematic of his ability to adapt to the changing landscape of cinema.

Fowley also made significant contributions to television, most notably as Doc Holliday in the popular ABC western series in the 1950s and early '60s. His television career was marked by versatility, as seen in his role as Grandpa Hanks in the CBS comedy in 1966-67.
