A big hello to the valued Life Style community! It's finally summer, my favorite season, which also coincides with the end of the semester and vacation time. However, these last few weeks of the semester are when things get more complicated, and this week has been truly tough. I had to coordinate a microbiology lab assignment, and oh my! Pulling that assignment off was a real challenge.

Sin embargo en estas últimas semanas que restan del semestre es cuando las cosas se vuelven más complicadas y precisamente esta semana ha sido realmente ruda, me ha tocado coordinar una práctica de laboratorio basada en microbiología y oh amigo! Llevar a buen término esa práctica fue una verdadera odisea.

The setup for this practice began a couple of weeks ago, when the days were rainy. I had to start preparing the solutions for this practice, which basically involves bacterial growth, that is, growing a strain of E. coli under different carbon sources. So the first step was to prepare physiological saline solution and then carbon sources such as glucose, galactose, succinate and lactose, which are the ones that this enterobacteria commonly feed on.

El montaje de esta práctica comenzó un par de semanas atrás, cuando los días eran lluviosos, tenía que comenzar preparando las soluciones de esta práctica que básicamente se trata de crecimiento bacteriano, es decir someter a una cepa de E. coli a crecer bajo distintas fuentes de carbono. Así que lo primero fue preparar solución salina fisiológica y posteriormente las fuentes de carbono como glucosa, galactosa, succinato y lactosa que son las que comúnmente se alimenta esta enterobacteria.

The aliquots of bacteria, the carbon sources, and the dilutions of the bacteria in saline solution must be performed using glass pipettes, which must also be sterilized to avoid contamination. This is one of the most time-consuming processes, as the different volumes of pipettes must be organized and then sterilized in the autoclave.

Las alícuotas de las bacterias, la toma de fuentes de carbono y las diluciones de las bacterias en solución salina se deben realizar con pipetas de vidrio que además deben esterilizarse para no contaminarse. Es uno de los procesos que más lleva tiempo, pues hay que organizar los distintos volúmenes de pipetas para posteriormente llevarlos a esterilizar en el autoclave.

Once all the materials to be used in the practice have been organized, they must be sterilized in an autoclave. This equipment necessarily uses distilled water so that it can evaporate in the equipment and generate the necessary pressure to sterilize all the glass and metal materials to be used. This is the longest part of the process because once sterilization is complete, the temperature and pressure must be allowed to drop before the material can be safely removed.

Una vez organizado todo el material que se utilizará en la práctica, éste debe llevarse a esterilizar en un autoclave. Es un equipo que utiliza agua destilada necesariamente para que pueda ser evaporada en el equipo y generar la presión necesaria para dejar estéril todo el material de vidrio y metal que se va a utilizar. Es la parte del proceso más largo ya que una vez que termina la esterilización, se debe esperar a que baje la temperatura y la presión para poder retirar el material de forma segura.

The entire process I explained above took two full weeks. When the day of the practice arrived, the students were evaluated with a quiz and then given a review of the protocol they must follow during the practice. I used this time to prepare the counters and ensure each team had all their sterile equipment. But just as we were about to begin, I received the terrible news that the main gas cylinder had to be shut off due to the risk of a leak. What a disaster!

Todo el proceso que te expuse anteriormente tomó 2 semanas completas, cuando llegó el día de realizar la práctica los estudiantes son evaluados con un quiz y posteriormente se les da una un repaso del protocolo que deben seguir en la práctica. Este es un momento que aprovecho para preparar los mesones y que cada equipo tenga todo su material estéril, pero justo cuando ya estábamos por comenzar, recibo la terrible noticia que se tuvo que cerrar la bombona principal de gas por riesgo de fuga, el desastre!

This was the worst-case scenario: no gas. How would it be possible to maintain a sterile environment using burners? Well, friends, that's where I used my inventiveness and creativity to save the day. The solution was to use alcohol burners to create a sterile environment and be able to inoculate the bacteria in a minimal medium with their specific carbon source and begin measuring growth indirectly by light scattering.

Este era el peor escenario posible, sin gas ¿Cómo sería posible mantener un entorno estéril usando los mecheros? bien amigos, ahí es donde usé la inventiva y la creatividad para salvar el día. La solución fue utilizar mecheros de alcohol para levantar un entorno estéril y poder realizar las inoculaciones de la bacteria en medio mínimo con su fuente de carbono específica y comenzar a medir el crecimiento de forma indirecta por dispersión de luz.

The greatest risk is when a volume of these bacteria must be taken and plated on an agar plate to count the bacterial colonies to estimate the bacterial growth rate, as they run the risk of contamination. These plates must remain incubated for 24 hours at 37°C in an oven. Fortunately, the students did their job well and their bacterial colonies grew in an appropriate quantity to be able to count them and estimate the growth.

El mayor riesgo es cuando debe tomarse un volumen de estas bacterias y sembrarlas en una placa de agar para contar las colonias bacterianas para estimar la tasa de crecimiento bacteriano, pues corren el riesgo de contaminarse . Estas placas deben permanecer por 24 horas incubadas a 37 °C en una estufa, afortunadamente los estudiantes hicieron bien su trabajo y les crecieron sus colonias bacterianas en cantidad apropiada para poder contarlas y estimar el crecimiento.

And so, friends, we managed to successfully complete the lab practice. I won't lie to you, I was so tired that day that I fell into bed exhausted until the next day. But there it is: the students successfully emulated the way Louis Pasteur did microbiology, and we saved the practice from being canceled due to a lack of gas.
