I remember that image of mTOR that you had. I thought I would use it and cite you, but was bit too focused on making a point about DNA repair. I might do that in my later posts when I revisit mTOR's role in others aspects of ageing.
I was aware of slightly aware of controversy regarding sirtuins. But I did not know that resveratol activation of sirtuins was an artifact of fluorophore. So thanks for pointing that out. I think that bothered me about sirtuins anyway is their role in promoting Non homologous end joining repair. On one hand where NHEJ is crucial for repairing double stranded breaks, too much of it can be detrimental for genome stability. This is also interesting because rapamycin causes suppression of DSB repair by NHEJ, while sirtuin activation might promote it. This is something I was saving for end of the series. So sirtuins slowing ageing is strange, perhaps a lot like it's dual role in cancer. Maybe unbdeerstading why it worked a certain times and why not at other times will yield a cleaner picture.
